Kahn was refering to his thoughts on materiality within a building.
"What a building wants to be" is basically about designing a building that is true to its function and the materials that it is made from.
Kahn said that he wasn't arbitrarily able to cut an opening in a brick wall for instance but he said to the brick what do you want to be - the brick said an arch so the building has arched openings
not sure if that quote above is right but its something along that line.
