Do you mean the thickness of the roof structure or of the growing medium/substrate?

The thickness of a wood or light metal framed roof shall be determined by the thickness of the insulation required by building code in the area where the building is erected.

Uniform and concentrated loads plus snow loads are an important factor to determine the depth (size) of the roof members such as joists and/or slabs.

Concrete slab roofs may not need additional insulation, the thickness of a concrete slab is determined by the distance between suporting points.

The compose/soil placed on the roof to grow grass or flowers, etc. is considered an insulating factor, trees are usualy plased in self contained vessels.

I've still got little idea of what you are asking for. As the previous poster says, the structural thickness will depend entirely on the method of construction of your building.

As for the roof garden, the depth of the substrate could be anything from 200mm for a grass roof to, I would say, 1 metre for supporting a small tree.

There are green roofs with thick soil (intensive) and with thin soil (extenstive).

This article talks about some benefits of the thicker type of green roof growing system:

This article is more about the thin type of roof...
